Structural elements of the Streptomyces oriC region and their interactions with the DnaA protein

Article Abstract:

The oriC region of the chromosome of three Streptomyces species were compared to determine the structural features of the replication origin. Results reveal that the oriC region of Streptomyces contains 19 conserved DnaA boxes. Plasmon resonance studies were conducted to characterize the binding interactions between these boxes and their binding proteins. Since the chromosome is resistant to unwinding using conventional procedures, there may be a yet uncharacterized protein that interacts with oriC to initiate unwinding and replication.

Transcription analysis of the dnaA gene and oriC region of the chromosome of Mycobacterium smegmatis and Mycobacterium bovis BCG, and its regulation by the DnaA protein

Article Abstract:

Transcriptional analysis in Mycobacterium smegmatis and M. bovis BCG demonstrates that the dnaA gene of M. bovis BCG is transcribed by two promoters, while trancription of dnaA gene in M. smegmatis is mediated by a single promoter. Furthermore, dnaA gene is autoregulated and expressed at all growth stages of the bacteria.

Inhibition of chromosome replication in Mycobacterium smegmatis: effect of the rpmH-dnaA promotor region

Article Abstract:

The rpmH-dnaA promotor region appears to inhibit the mycobacterial origin of replication oriC in Mycobacterium smegmatis. Plasmids that contain the origin function adjacent to the dnaA gene or its regulatory region do not yield transformants.
