The extracellular acid protease gene of Yarrowia lipolytica: sequence and pH-regulated transcription

Article Abstract:

The regulation of expression of the acid protease (AXP) gene of Yarrowia lipolytica 148 was investigated by molecular methods. Isolated DNA from AXP consisted of 353 amino acids. Sequencing of the AXP gene revealed a putative 17 amino acid hydrophobic prepeptide and a 27 amino acid propeptide which lack N-glycosylation sites. Cleavage between residues Phe and Ala induced enzyme synthesis. Northern blotting analysis confirmed AXP as a member of the aspartyl family of proteases that shares homology to proteases of a number of fungi and of human progastrin.

Dipeptidyl aminopeptidase processing and biosynthesis of alkaline extracellular protease from Yarrowia lipolytica

Article Abstract:

A research on alkaline extracellular protease (AEP) that comes from Yarrowia lipolytica was conducted to determine its aspects in processing and secretion, the site of signal peptide cleavage, and the role of the dipeptide stretch and DPAPase processing in AEP maturation. Analysis of yeast strain, In vitro mutagenesis, and protease assays were the primary methodology used. Findings showed that the inhibition of DPAPase activity allow secretion of mature-sized-AEP. Dipeptide stretch was also found to be important in the secretion of heterologous proteins.

pH-regulated expression of the acid and alkaline extracellular proteases of Yarrowia lipolytica

Article Abstract:

A study was conducted on the pH-regulated expression of the acid (AXP) and alkaline extracellular proteases (AEP) of Yarrowia lipolytica. Northern blotting, enzyme assays and Western blotting were used to determine the expression in batch and continuous cultures at the mRNA level. Results have shown that the highest amounts of AEP mRNA occur at pH 6.5, while the highest amounts of AXP mRNA occur at pH 5.5.
