The polyadenylation factor CstF-64 regulates alternative processing of IgM heavy chain pre-mRNA during B cell differentiation

Article Abstract:

The influence of transacting factors in mu gene regulation is investigated. The polyadenylation factor CstF-64 is shown to be specifically up-regulated during B cell differentiation. Overexpression of the factor in a B cell line raises intact CstF concentration and reproduces the shift in IgM pre-mRNA processing that occurs during differentiation. A detailed model for regulating the mum-to-mus mRNA switch is also proposed.

The upstream sequence element of the C2 complement poly(A) signal activates mRNA 3' end formation by two distinct mechanisms

Article Abstract:

The C2 complement gene's poly(A) signal is atypical. It has a upstream sequence element (USE) necessary for complete activity in vivo. The upstream sequence part of the C2 complement poly(A) signal activates mRNA 3' end formation via two separate mechanisms. An interconnection exists between spicing and polyadenylation as does unexpected flexibility in the organization of mammalian poly(A) sites.

Symplekin and xGLD-2 are required for CPEB-mediated cytoplasmic polyadenylation

Article Abstract:

Two new factors, symplekin, a binding protein and xGLD-2, an unusual poly(A) polymerase that are essential for polyadenylation are defined. The identification of these factors has broad implication for biological processes that employ polyadenylation-regulated translation, such as gametogenesis, cell cycle progression, and synaptic plasticity.
