Three-dimensional structure of an evolutionarily conserved N-terminal domain of syntaxin 1A

Article Abstract:

Analysis of the three-dimensional structure of the N-terminal region of syntaxin 1A using nuclear magnetic resonance spectroscopy indicates that residues 27-146 constitute an independently folded domain with a three-helix bundle structure. A long groove is formed by the interface between the second and third helices. This groove shows a strong residue conservation in plasma membrane synataxins and probably provides a binding site for an alpha-helix of another protein of the exocytotic machinery. Further research into which proteins interact directly with the groove could produce interesting results.

A complexin/synaptotagmin 1 switch controls fast synaptic vesicle exocytosis

Article Abstract:

A study was conducted to demonstrate that complexin binding activates SNARE complexes into a metastable state and that C[a.sup.2+] binding to synaptotagmin 1 triggers fast exocytosis by displacing complexin from metastable SNARE complexes. The results state that complexins activate vesicles by converting SNARE complexes from a loose to a tightly assembled state that serves as essential substrate for subsequent synaptotagmin 1 function.

Otoferlin, defective in a human deafness form, is essential for exocytosis at the auditory ribbon synapse

Article Abstract:

Otoferlin, a predicted C2-domain transmembrane protein, which is defective in a recessive form of human deafness, is studied. It shows that otoferlin expression in the hair cells correlates with afferent synaptogenesis and otoferlin localizes to ribbon-associated synaptic vesicles.
