Zymomonas mobilis squalene-hopene cyclase gene (shc): cloning, DNA sequence analysis, and expression in Escherichia coli

Article Abstract:

The use of an Alicyclobacillus acidocaldarius squalene-hopene cyclase (shc) gene-derived DNA probe helps clone a Zymomonas mobilis HindIII chromosomal DNA piece that is 4.3 kb long. A 1977 bp open reading frame codes for a 658-amino acid protein of 74 kDa molecular mass. shc expression in Escherichia coli K12 strains is regulated by lac or tac promoters, with the product exhibiting squalene-hopene cyclase function. Sequence alignments with Candida albicans canosterol cyclase, Arabidopsis thalina cycloartenol synthase and A. acidocaldarius SHC reveal the presence of six Gln-X-X-X-Gly-X-Trp core group-containing conserved regions in the proteins.

Cloning, sequence analysis, expression and inactivation of the Corynebacterium glutamicum pta-ack operon encoding phosphotransacetylase and acetate kinase

Article Abstract:

A study was conducted to analyze the cloning, expression and inactivation of the Corynebacterium glutamicum pta-ack operon encoding acetate kinase and phosphotransacetylase. Results indicated no isoenzymes of acetate kinase and phosphotransacetylase present in Corynebacterium glutamicum. Findings also showed the importance of a functional acetate kinase/phosphotransacetylase pathway in the growth of the organism on acetate.

Squalene-hopene cyclase from Bradyrhizobium japonicum: cloning, expression, sequence analysis and comparison to other triterpenoid cyclases

Article Abstract:

The cloning, DNA sequencing, expression and sequence analysis of the squalene-hopene cyclase (SHC) gene from Bradyrhizobium japonicum USDA 110 was described. A two-step PCR-based screening method was used to isolate the SHC gene. The B. japonicum gene was expressed in Escherichia coli and was found similar to eukaryoric oxidosqualene cyclases. SHC and ORF were suggested as parts of an operon due to their close proximity.
