Article Abstract:

The structure and location of protein in living cells is studied using a modular labeling method, where the perturbations caused to the concerned protein at the molecular level is minimized. Nitrilotriacetate (NTA) and fluorescein arsenical hairpin approaches are complementary and can be used in combination for determining the intramolecular fluorescence resonance energy transfer (FRET) with well-defined localization.

A method for selecting aggregation-resistant proteins by heat denaturation is described. This method is illustrated with antibody heavy chain variable domains, which are prone to aggregate.
