Dependence of tryptophan emission wavelength on conformation in cyclic hexapeptides

Article Abstract:

Quantum mechanical/molecular dynamics (QM/MD) simulations of the six canonical side chain rotamers of tryptophan in the seven cyclic hexapeptides are described. The emission maxima of the three decay-associated spectra for the seven peptides ranged form 314 to 359nm, suggesting that different tryptophan rotamers have different emission maxima even in the case of solvent-exposed tryptophans and these spectra could aid in the assignment of fluorescence lifetimes to tryptophan rotamers.

Quantitative prediction of fluorescence quantum yields for tryptophan in proteins

Article Abstract:

A reasonable basis for the 30-fold variation by the use of quantum mechanics-molecular mechanics simulations is proposed in which the energy of the lowest Trp ring-to-amide backbone charge transfer (CT) state is monitored during dynamics trajectories for 24 Trps in 17 proteins. The full range of tryptophan (trp) fluorescence quantum yields observed in proteins is due primarily to different degrees of electron transfer from the indole ring to a local amide.
