Simplified hot start PCR

Article Abstract:

Polymerase chain reaction (PCR) with thermally activated DNA polymerase gives greater product yield, and is more sensitive and specific. AmpliTaq Gold DNA polymerase used for PCR does not show any enzyme activity at room temperature but can be substituted in most PCR systems for thermostable DNA polymerase. A pre-PCR heat cycle restores the polymerase activity, and is programmable such that the conversion from enzyme activation to PCR cycling activity is above the optimal primer annealing temperature.
