Treatment of carrageenan induced arthritis by the platelet activating factor antagonist BN 50730

Article Abstract:

Platelet activating factor (PAF) may be part of the development of early stage arthritis. PAF is released by various inflammatory cells and activates two types of white blood cells involved in immune responses. Increased concentrations have been found in patients with active rheumatoid arthritis. Arthritis was induced in the knee of rabbits by injecting carrageenan weekly for five weeks. Some rabbits were also treated with a PAF receptor antagonist at the time of each injection, which would blunt the formation of PAF. Others were treated with the PAF receptor antagonist beginning on day 28, after the inflammatory process was underway. Yet other rabbits received no receptor antagonist. On day 60 animals were killed and knee tissue was microscopically analyzed. Animals treated simultaneously with the antagonist had lower arthritis scores than untreated animals. Arthritis scores in animals treated after a lag time dropped to that of levels of rabbits treated simultaneously. Therefore, the suppression of PAF inhibited the arthritic inflammation process.

Clinical and laboratory manifestations of elderly onset psoriatic arthritis: a comparison with younger onset disease

Article Abstract:

Psoriatic arthritis seems to begin and develop much more aggressively in older people over 60 years of age than it does in younger patients. Analysis of synovial fluid of both patients with elderly onset psoriatic arthritis (EOPsA) and younger onset psoriatic arthritis (YOPsA) showed higher levels of Interleukins such as IL1Beta, IL6, and IL8. These concentrations suggest that immune system changes in the EOPsA are responsible for the intensity of onset with those people.
