Inhibition of cytotoxicity and cytokine release of CD8+ HIV-specific cytotoxic T lymphocytes by pentoxifylline

Article Abstract:

The anti-inflammatory drug pentoxifylline (POX) may inhibit HIV replication by suppressing the production of cytokines by CD8+ cytotoxic T lymphocytes (CTLs). Cytokines are naturally-occurring immune system chemicals such as interferons, tumor necrosis factor and interleukins. Researchers incubated CD8+ CTLs specific to HIV with a human B cell line and measured the ability of the CTLs to destroy the B cells. The cytotoxic activity of the CTLs was inhibited by POX. In addition, POX inhibited the release of interferon-gamma, tumor necrosis factor-alpha and granulocyte-macrophage colony stimulating factor by the CTLs. This could be important in the treatment of HIV infection because tumor necrosis factor-alpha is known to enhance HIV replication. On the other hand, suppression of the cytotoxic activity of CTLs could allow the virus to flourish.

Induction of HIV-1 replication in a chronically infected T-cell line by cytotoxic T lymphocytes

Article Abstract:

Activation of HIV-1-specific cytotoxic T cells (CTL) may induce replication of HIV type 1 (HIV-1) in chronically infected T cells. CTLs are cells of the immune system that can recognize and lyse target cells with specific types of antigens on their surface. A study examined the effect of HIV-1-specific CTLs on HIV-1 replication in chronically infected T cells in the laboratory. Activation of CTLs with subunits of protein induced replication of HIV-1. Increased replication of HIV-1 was associated with elevated production of tumor necrosis factor-alpha (TNF-alpha) by CTLs. Antibodies against TNF-alpha could inhibit the effect of activated CTLs on HIV-1 replication.

Rapid and selective depletion of CD4+T lymphocytes and preferential loss of memory cells on interaction of mononuclear cells with HIV-1 glycoprotein-expressing cells

Article Abstract:

This study looks at the death of uninfected CD4+T cells and seeks the cause of it. It observes that it might be the same as that of glycoprotein-expressing cells with CD4+T lymphocytes.
