Role of plasminogen-activator inhibitor type 1 in the pathogenesis and outcome of the hemolytic uremic syndrome

Article Abstract:

Children with hemolytic uremic syndrome may have increased blood levels of plasminogen-activator inhibitor type 1 (PAI-1), an enzyme that inhibits the break up of fibrin. Hemolytic uremic syndrome is a kidney disorder characterized by the deposition of fibrin, a protein involved blood clotting, in kidney blood vessels. Blood levels of PAI-1 in 40 children hospitalized for hemolytic uremic syndrome were higher than those of nine children with other types of kidney failure. Twelve children with hemolytic uremic syndrome regained kidney function without dialysis, 23 regained kidney function after dialysis and five did not regain kidney function after dialysis. Blood levels of PAI-1 were elevated upon admission to the hospital in these children, but decreased significantly after dialysis. Severity of disease was associated with the amount of time blood levels were PAI-1 were elevated.

Plasminogen-activator inhibitor type 1 and coronary artery disease

Article Abstract:

Plasminogen-activator inhibitor type 1 (PAI-1) may play a role in coronary artery disease. PAI-1 inhibits tissue plasminogen activator (t-PA), an enzyme that dissolves blood clots. This prevents blood clots from being eliminated, which could increase the risk of a heart attack if the clots are in the coronary arteries. People with high blood levels of PAI-1 have a higher risk of abnormal blood clotting and other diseases involving blood vessels.

Glutathione peroxidase 1 activity and cardiovascular events in patients with coronary artery disease

Article Abstract:

Low blood levels of an enzyme called glutathione peroxidase 1 may be a risk factor for coronary artery disease, according to a study of 636 patients. Those with the highest blood levels of this enzyme were about 80% less likely to develop coronary artery disease than those with the lowest blood levels of the enzyme. Glutathione peroxidase 1 is an antioxidant enzyme that eliminates free radicals.
