Human cytochromes P450: problems and prospects

Article Abstract:

Research on human P450 cytochromes will have an important effect on drug administration and dosage and on public health and research. P450s are responsible for the matabolism of foreign compounds in the body. Genetic and racial differences in P450s makes pharmacogenetics difficult. In addition, human P450s have evolved differently from the P450s of animals, making experimentation on animals difficult. New systems need to be developed to effectively study and control P450 levels in humans.

Diagnostic analysis, clinical importance and molecular basis of dihydropyrimidine dehydrogenase deficiency

Article Abstract:

Dihydropyrimidine dehydrogenase (DPD) is a cytosolic enzyme that is present in the three-step catabolic pathway of thymine and uracil, which is necessary for the synthesis of amino acid. It was extensively used in the production of anticancer drugs, but resulted to high toxicity among patients. Individuals deficient in DPD have low heterozygote frequency and exhibit phenotypic abnormalities. However, there is still no standard procedure for diagnosing DPD-deficient patients.

Inhibitory monoclonal antibodies to human cytochrome P450 enzymes: a new avenue for drug discovery

Article Abstract:

Monoclonal antibodies that react with cytochrome P450 can be used to determine which isoform of the enzyme an individual has. This in turn would determine how that individual would metabolize certain drugs.
