A general mechanism for transcriptional synergy by eukaryotic activators

Article Abstract:

The DNase I footprint and gel shift assays show a correlation between the synergistic transcriptional effect of promoter sites and activation modules and the TFIID:TFIIA (DA) complex. The synergism degree of the activator-dependent DA is enhanced by TFIIB when their interaction is regulated by TATA-binding protein-associated factors, which was similar for acidic activator GAL4-VP16 but different than the basal DA complex. A unifying mechanism for gene activation and synergy by acidic and non-acidic activators has been suggested.

The advantage of sex in evolving yeast populations

Article Abstract:

Researchers have looked at the impact of sex on mean fitness in experimental populations of the budding yeast Saccharomyces cerevisiae. It was found that sex boosts mean fitness in circumstances to which populations have become well adapted. This seems to back up the assumption that sexual activity in eukaryotes serves mainly to remove negative mutations. Sex does not speed up the rate at which yeast populations become adapted to a new environment.

Binding of TBP to promoters in vivo is stimulated by activators and requires Pol II holoenzyme

Article Abstract:

Transcriptional activators may act by recruiting the RNA polymerase II (Pol II) machinery in eukaryotes, by changing the conformation of the machinery. The TATA-box-binding-protein (TBP) occupancy of 30 yeast promoters has been studied in vivo. TBP association with promoters is found to be stimulated by activators and restricted by the Cyc8-Tup1 repressor.
