Androstane metabolites bind to and deactivate the nuclear receptor CAR-beta

Article Abstract:

The DNA binding orphan receptor CAR-beta acts as a heterodimer with the retinoid-X receptor, activating gene transcription in a constitutive way. The constitute activity of CAR-beta is shown to result from a ligand-independent recruitment of transcriptional co-activators. The steroids androstanol and androstenol were found to inhibit constitutive activity of CAR-beta, but do not interfere with the heterodimerization or DNA binding of CAR-beta.

Interaction of thyroid-hormone receptor with a conserved transcriptional mediator

Article Abstract:

Experimental studies of a yeast two-hybrid system suggest the ability of the thyroid-hormone-receptor interacting protein (Trip1), which possesses structural homology with the transcriptional mediator Sug1, to react in vitro with the regulatory protein GAL4's activation domain and with the thyroid-hormone receptor. This implies a role of Trip1 as a transcription activator in yeast.
