Budding from Golgi membranes requires the coatomer complex of non-clathrin coat proteins

Article Abstract:

The role of coat proteins in Golgi membrane budding was investigated. The membrane-mediated budding hypothesis states that budding is an intrinsic property of Golgi membranes which can thus proceed without extrinsic coat proteins. On the other hand, the coat-mediated budding hypothesis states that coat proteins are essential for budding to occur. The study involved a cell free system in which coat proteins formed from Golgi cisternae are immunodepleted. The results proved that coat proteins are essential for Golgi membrane budding.

Inhibition by brefeldin A of a Golgi membrane enzyme that catalyses exchange of guanine nucleotide bound to ARF

Article Abstract:

A new Golgi membrane enzyme that assists in the guanine nucleotide (GDP-GTP) exchange within the adenosine diphosphate-ribosylation factor has been identified through the ability of brefeldin A (BFA), a fungal metabolite, to block the exchange. BFA's inhibitory effect is strong evidence for an enzymatic nucleotide swapping device that is necessary for the development of budding vesicles.

Brefeldin A inhibits Golgi membrane-catalysed exchange of guanine nucleotide onto ARF protein

Article Abstract:

Brefeldin A (BRA), a fungal metabolite, disrupts the Golgi membrane-controlled catalysis by which GTP-binding protein is swapped for adenosine diphosphate-ribosylation factor (ARF). BRA effects this disruption by binding to guanine nucleotide. BRA's inhibitory property suggests the existence of a Golgi-linked binding protein whose chief function is to link ARF with guanine nucleotide.
