Crystal structure of a bacteriophage T7 DNA replication complex at 2.2A resolution

Article Abstract:

Polymerases, working in association with accessory proteins unwinding the DNA duplex, replicate genomic DNA. A 2.2A crystal structure of the replicative DNA polymerase from bacteriophage T7 is presented, complexed with a primer-template and nucleoside triphosphate in the polymerase active site. It shows how nucleotides are chosen in a template-directed manner and gives a structural base for metal-assisted phosphoryl transfer mechanism.

CDK-dependent phosphorylation of Sld2 and Sld3 initiates DNA replication in budding yeast

Article Abstract:

A study was conducted to show that both an allele of CDG45(JET1) and high-copy DPB11, in combination with Sld2-11D, separately confer cyclin-dependent kinase (CDK)-independent DNA replication. The results demonstrate that CDKs regulate interactions between BRCT-domain-containing replication proteins and other phosphorylated proteins for the initiation of chromosomal DNA replication.

DNA primase acts as a molecular brake in DNA replication

Article Abstract:

The kinetics of leading- and lagging-strand synthesis at the single-molecule level by monitoring the length of individual DNA molecules during the replication reaction is characterized. It is observed that the synthesis of primers on the lagging strand to cause transient pausing of the highly processive leading-strand synthesis.
