Crystal structure of a hepatitis delta virus ribozyme

Article Abstract:

The hepatitis delta virus (HDV) ribozyme is a catalytic RNA required for the viability of a human pathogen. Crystals of a 72-nucleotide, self-cleaved form of the genomic HDV ribozyme were obtained. The crystal structure has a compact catalytic core consisting of five helical segments linked as a pseudoknot. The self-scission reaction results in the 5'-hydroxyl leaving group buried within an active-site cleft, surrounded by biochemically significant backbone and base functional groups.

A conformational switch controls hepatitis delta virus ribozyme catalysis

Article Abstract:

Ten crystal structures of the hepatitis delta virus (HDV) ribozyme in its pre-cleaved state, showing that cytidine are positioned to activate the 2'-OH nucleophile in the precursor structure is reported. The observation implies that RNA structural rearrangements control the reactivity of ribozymes and ribonucleoprotein enzymes.

Crystal structure of a hairpin ribozyme-inhibitor complex with implications for catalysis

Research is presented concerning the structure of the hairpin ribozyme-inhibitory complex. The role of the hairpin ribozyme in the catalysis of RNA sequence-specific cleavage is discussed.
