Calcium signalling in the guidance of nerve growth by netrin-1

Article Abstract:

It has been established that Ca2+ influx and Ca2+-induced Ca2+ release (CICR) play a role in mediating growth-cone guidance induced by the guidance molecule netrin-1. The netrin-1 induced turning response depends on Ca2+ influx through plasma membrane Ca2+ channels and on Ca2+-induced Ca2+ release from cytoplasmic stores. This research indicates the significance of the precise profile of Ca2+ gradients in establishing the turning response of the growth cone.

Cyclic AMP/GMP-dependent modulation of Ca (super)2+ channels sets the polarity of nerve growth-cone turning

Article Abstract:

Research has been conducted on cyclic AMP and GMP. The authors demonstrate that cyclic AMP to cyclic GMP activity ratio affects netrin-1-induced axon guidance polarity.

Growth-cone attraction to netrin-1 is converted to repulsion by laminin-1

Article Abstract:

Extracellular matrix (ECM) molecules modulate the response of growing axons to secreted guidance cues, as well as acting in substrate-mediated adhesion. It has been established that, in Xenopus, laminin-1 from the ECM converts netrin-mediated attraction into repulsion. The netrin-1-induced rise in cyclic AMP could assist in promoting a polymerization-based attractive response, while laminin-1 could encourage a repulsive response by favouring netrin-1-mediated depolymerization.
