DREAM is a Ca2+-regulated transcriptional repressor

Article Abstract:

A human caudate expression library has been screened using a double-stranded downstream regulatory element (DRE) oligonucleotide as a probe in order to isolate complementary DNAs encoding proteins capable of binding to the DRE sequence. It was possible to isolate DRE-agonist modulator (DREAM), a new transcriptional repressor. DREAM, which has four CA2+-binding domains of the EF-hand type, specifically binds to the DRE. This seems to be the first time that it has been shown that a nuclear Ca2+ sensor directly represses transcription by binding specifically to DNA.

Spreading of transcriptional repressor SIR3 from telomeric heterochromatin

Article Abstract:

The SIR3 transcriptional repressor is a structural component of Saccharomyces cervisiae heterochromatin that suppresses the transcription of adjacent genes as it spreads along the chromosome. SIR3 co-immunoprecipitates SIR4, RAP1 and histones from cellular extracts. This suggests that large chromatin-related protein complexes are present in cells. SIR3 is also present at HMRa, HML-alpha and telomeres in vivo. When overexpressed, it spreads from the telomeric regions into the adjacent chromatin.

Paradox of a tumour repressor

Article Abstract:

The application of the copies of chromosome 21 to reduce the presence of solid tumours in humans is discussed. The chromosomes reduce the tumours, though they have a high risk of initiating cancer metastasis in the patients.
