Formation of haematopoietic microenvironment and haematopoietic stem cells from single human bone marrow stem cells

Article Abstract:

Two different subsets of pluripotent stem cells taken from human fetal bone marrow show distinct qualities. One can differentiate into all hematopoietic lineages, the other into both hematopoietic precursors and stromal cells. The stromal cells are capable of helping the differentiation of the precursors. This is the first time that a single cell has been linked to the reconstitution of both hematopoietic cells and the bone marrow microenvironment which is associated with them. These results suggest that gene therapy might be most profitably directed at purified common stem cells.

Inefficient positive selection of T cells directed by haematopoietic cells

Article Abstract:

Class I major histocompatibility complex (MHC) molecules expressed by hematopoietic cell types may control the positive selection of CD8+ T cells but at a slower rate than is effected by thymic epithelial cells. This finding directly contradicts earlier research indicating that the MHC molecules expressed on non-hematopoietic cells control the positive selection. The nature of the cell types that regulate selection will strongly influence attempts to construct mechanistic models of how T cells are selected.

PTEN maintains haematopoietic stem cells and acts in lineage choice and leukaemia prevention

Article Abstract:

A demonstration that activation of phosphatase and tensin homologue (PTEN) in bone marrow Haematopoietic stem cells (HSCs) causes their short-term expansion, but long-term decline, primarily owing to an enhanced level of HSC activation is presented. HSC must achieve a balance between quiescence and activation that fulfils immediate demands for haematopoiesis without compromising long-term stem cell maintenance.
