In vitro development of zygotes from prepubertal gilts after microinjection of DNA

Article Abstract:

A study was done to evaluate the culture procedures and incubation procedures for DNA-microinjected zygotes. The purpose was to understand why transgenic animal experiments are hampered by inadequate in vitro development of zygotes. Precocious ovulation in prepubertal gilts was induced by treatment with human choriogonadotropin. Zygotes received a pronuclear microinjection of buffer, buffer with DNA construct, or no microinjection. The results showed that zygotes microinjected with DNA had a lower cleavage index, indicating that DNA microinjection is detrimental to zygote development.

Insulin-like growth factors and binding proteins in ruminants and their nutritional regulation

Article Abstract:

Insulin-like growth factors (IGF) influence growth, reproduction and other health aspects, in ruminants. The exact mechanism of action of IGF is not known though it is believed to be influenced by other growth factors and state of development of the cell. Somatomedins and related binding proteins act as a link between cellular development and supply of nutrients. In ruminants, the concentration of circulating IGF is decreased under conditions of severe diet restriction. Even moderate changes in the nutritional status affect IGF response to somatotropins.
