Linking a genetic defect to its cellular phenotype in a cardiac arrhythmia

Article Abstract:

Genetic defects in membrane ion channels can upset the delicate balance of dynamic interactions between ion channels and the cellular environment, resulting in altered cell function. A single-channel-based Markovian modelling approach is described to establish a connection between molecular findings and the physiology and pathophysiology of the cell. The cellular arrhythmogenic consequences of a mutation in the cardiac sodium channel that can result in a clinical arrhythmogenic disorder and sudden cardiac death is determined.

Enzymatic activation of voltage-gated potassium channels

Article Abstract:

The activation of voltage-gated [K.sup.+] (Kv) channels by a sphingomyelinase is reported. This has provided significant insight into the way cell excitability could be regulated by altering the properties of lipids in the plasma membrane and the way voltage-gated channels could be activated in non-excitable cells or lower organisms.

hERG potassium channels and cardiac arrhythmia

Article Abstract:

hERG potassium channels are essential for normal electrical activity in the heart. Arrhythmia is induced by a blockage of hERG channels by a diverse group of drugs, and insights gained from the crystal structures of other potassium channels help understand the block of hERG channels and the mechanisms of gating.
