Oligomerization activates c-Raf-1 through a Ras-dependent mechanism

Article Abstract:

The oligomerization of the c-Raf-1 proto-oncoprotein by a Ras-dependent mechanism facilitates Raf activation. The addition of FK506 derivatives of FK1012A induces the oligomerization of recombinant FKBP-Raf in situ, which stimulates Raf kinase activity almost half as well as the epidermal growth factor. This activation of FKBP-Raf is dependent upon Ras-GTP, which translocates c-Raf-1 to the membrane, initiating an alteration that facilitates the interaction of the Raf polypeptide with an activator.

A dimeric 14-3-3 protein is an essential cofactor for Raf kinase activity

Article Abstract:

The cRaf-1 protein kinases is mitogen-activated and is the main effector used by GTP-bound Ras to activate the MAP kinase pathway. GTP bound Ras can bind Raf but is not sufficient for the stable activation of Raf. The role of 14-3-3 proteins in the regulation of Raf's kinase activity is investigated. It is proposed that dimeric 14-3-3 is required to maintain Raf in the inactive state in the absence of GTP-bound Ras, and to stabilize active Raf.

A signal for Beta-cell failure

Article Abstract:

Human type 2 diabetes is usually accompanied by insulin response defects, resulting in hyperglycaemia. Withers and colleagues have reported the phenotype of mice lacking alleles encoding the insulin-receptor substrate protein-2, IRS-2. The mice develop a syndrome like the common form of human type 2 diabetes.
