Pre-bending of a promoter sequence enhances affinity for the TATA-binding factor

Article Abstract:

Competition experiments reveal that binding affinity of TATA-binding protein (TBP) to DNA pre-bent towards the major groove is 100 times higher than the binding affinity of TBP to unbent DNA, and 300 times higher than the binding affinity of TBP to DNA pre-bent towards the minor groove. Various factors including chromatin in cells influence the bending in DNA, and changes in the affinity of the TATA factor by bending are vital for transcriptional regulation.

MAP kinase links the transcription factor Microphthalmia to c-Kit signalling in melanocytes

Article Abstract:

Microphthalmia (Mi) is a transcription factor of the basic, helix-loop-helix, leucine-zipper (bHLHzip) type, mutations of which can lead to Waardenburg Syndrome type II. It is suggested that Sl, c-Kit and Mi function with a common growth or differentiation pathway. A human melanoma cell line was studied using Western blot analysis, revealing two Mi species having mobilities corresponding to molecular masses of 54K and 60K.
