Shiga-like toxins are neutralized by tailored multivalent carbohydrate ligands

Article Abstract:

It has been possible to use the crystal structure of the B5 subunit of Escherichia coli 0157:H7 Shiga-like toxin I (SLT-I) in complex with an analogue of its carbohydrate receptor to design an oligovalent, water-soluble carbohydrate ligand. This ligand, designated STARFISH, has subnanomolar inhibitory activity. The STARFISH inhibitor was shown to protect susceptible cells against prolonged exposure to SLT-I and to the apparently more clinically significant SLT-II, a toxin for which there were previously no inhibitors.

Two crystal structures of Dpo4, an archael pol eta homologue, are determined, which are complexed with CPD-containing DNA, where the 3' and 5' thymine of the cyclobutane pyrimidine dimers (CPD) separately serves as a templating base.
