Telomeric tethers

Article Abstract:

It is possible that the three-dimensional organization of chromosomes is partly provided by interactions between nucleoporins and specific chromosomal regions. This research identified a connection between Ku proteins, telomere localization and nuclear pore complexes. It is suggested that Mlp proteins are responsible for the peripheral localization of telomeres, while Mlps are docked to the nuclear periphery by interaction with Nup145p. Future research on Mlps should produce more information about the link between telomeric silencing, DNA repair and telomere localization.

The yeast Pif1p helicase removes telomerase from telomeric DNA

Article Abstract:

The in vitro Pif1p reduces the processivity of telomerase and releases telomerase from telomeric oligonucleotide. The released telomerase is processivity active is able to lengthen a challenger oligonucleotide.

Control of telomere length by the human telomeric protein TRF1

Article Abstract:

The role of the human telomeric-repeating binding factor TRF1 in the regulation of telomere length is investigated. The impact of extended overexpression of wild-type TRF1 and of a dominant-negative mutant on telomere length in a telomerase-positive human tumor-cell line with stable telomeres is analyzed. Results show that TRF1 is a suppressor of telomere elongation and indicate that the enzyme has a significant role in the negative feedback mechanism stabilizing telomere length.
