Thermodynamic beta-sheet propensities measured using a zinc-finger host peptide

Article Abstract:

The thermodynamic beta-sheet propensities of 20 common amino acids are measured using a zinc-finger host peptide. The amino acids were substituted into a guest site of the host system which were unfolded without bound metal. The propensities of each substance was determined using a competitive cobalt(II)-binding assay. The resulting scale indicated the thermodynamic origin of beta-sheet propensities.

It has been possible to design and test new antibiotics that selectively disrupt the cell membranes of target microorganisms. These antibiotics are based on a cyclic peptide structure, with the synthetic ring peptides having the advantage of being significantly smaller than the alpha-helical ones, thus boosting their ability to penetrate tissues.
