Homologous gene replacement in Physarum

Article Abstract:

A new method facilitates the gene replacement in the protist Physarum polycephalum and thereby helps investigate cellular and developmental biology of this organism. Substitution of ardD-Delta-1 for ardD+ produces transformants, in which the vital carboxy-terminal region of the gene products is mutated by ardD-Delta-1. Growth and viability of different cell types of Physarum are not affected by ardD-Delta-1, suggesting that the carboxy-terminal region of the ardD product is not essential for the growth.

Molecular characterization of lengthy mitochondrial DNA duplications from the parasitic nematode Romanomermis culicivorax

Article Abstract:

The complete nucleotide sequences of 3.0-kilobase mitochondrial DNA repeating units isolated from isofemale lineages of the nematode Romanomermis culicivorax were reported. Analysis showed that each amplified unit contains the NAHD dehydrogenase subunits 3 and 6 genes, an open reading frame (ORF1) for a cytochrome p450-like gene, and three unidentified ORFs. In addition, the study showed that primary nucleotide sequences of the repeat copies are highly conserved.

A genetic system controlling mitochondrial fusion in the slime mould, Physarum polycephalum

Article Abstract:

A 16 kilobase pair mitochondrial plasmid was isolated from Physarum polycephalum individuals that exhibit mitochondrial fusion. This fragment, inserted in the mitochondrial genome of progenies of crosses between heterologous strains, appears to promote mitochondrial fusion in the slime mold. Further, genetic analysis of the progenies of crosses between three isolates reveled a number ofnuclear allelic loci regulating mitochondrial fusion.
