Strain typing with IS200 fingerprints in Salmonella abortusovis

Article Abstract:

Hybridization profiles of the IS200 insertion element of Salmonella abortusovis strains isolated from sheep reveal a band at almost 9 kb, besides additional strain-specific bands. The isolates from Sardinia, Italy, show low polymorphism and three IS200 hybridization bands. Most of the strains contain a copy of IS200 in the variable area of their virulence plasmid. Strain typing using IS200 fingerprinting is more accurate than chromosome restriction fragment length polymorphism typing, plasmid profiles and rRNA restriction fragment length polymorphism analysis.

Identification of Salmonella abortusovis by PCR amplification of a serovar-specific IS200 element

Article Abstract:

A polymerase chain reaction assay was developed for the identification of Salmonella abortusovis DNA based on the presence of the serovar-specific IS200 copy on the bacterial chromosome. The serovar-specific IS200 element in Salmonella abortusovis was detected in the 900-bp and 2-kb fragment by Southern hybridization against plasmids pIZ46 and pIZ47. Furthermore, analysis of the fragments from primer combinations indicated the presence of an extra DNA that is located on one side of the IS200 element.

Cost-effective application of pulsed-field gel electrophoresis to typing of Salmonella enterica serovar Typhimurium

Article Abstract:

The relationship between phage type and pulsed-field gel electrophoresis (PFGE) pattern in Salmonella enterica serovar Typhimurium isolates from humans, animals and food in Ireland is explored to determine how to apply PFGE to supplement the existing routine phenotypic subtyping of serovar Typhimurium. The analysis facilitates the selective application of PFGE to maximize the yield of epidemiologically relevant information while controlling costs.
