The regulation of E2F by pRB-family proteins
Article Abstract:
The E2F transcription factor and the product of the retinoblastoma tumor suppressor gene (pRB) represent families of related proteins with varied activities sometimes in opposition. E2F is important in regulation of cell proliferation in Drosophila, mammalian cells and likely other eukaryotes. E2F-1 acts as a standard oncogene in transformation assays. Its overexpression in transgenic mice pushes tumorigenesis. Its absence cuts down on tumor phenotypes from inactivation of pRB. E2F-1 can be both an oncogene and tumor suppressor gene.

p53: Link to the past, bridge to the future
Article Abstract:
The presently available molecular techniques are used to revisit the structure of p53, the founding member of this gene family that plays a pivotal role in human cancer. A comparison of p53-related genes reveals strikingly similar traits among family members that produce a plethora of related but functionally diverse proteins that add further complexity to the role of p53-related proteins in development, cancer, and aging.

Identification of a novel class of genomic DNA-binding sites suggests a mechanism for selectivity in target gene activation by the tumor suppressor protein p53
Article Abstract:
Two response elements exist for p53 in the promoter of the cyclin-dependent kinase inhibitor p21 gene. Analysis using mutation has indicated that a single-base change brought about behavior of one element like that of the other. Two classes of p53-binding sites have been identified. It appears that there is a mechanism for target gene selectivity by p53.
