Activation of topoisomerase II-mediated excision of chromosomal DNA loops during oxidative stress
Article Abstract:
Topoisomerase II (TOP2) potentially has a role in oxidative damage of DNA and cell death that is apoptotic. Hydrogen peroxide is known to bring on oxidative streess and apoptosis and U937 cells treated with it have been shown to produce high molecular weight DNA fragments from about 50 to 100 kb in less than a minute. This fragment formation is reversible and mediated by TOP2. After the first event, irreversible high molecular weight DNA fragments and nucleosomal ladders are formed.

Suppression of the p300-dependent mdm2 negative-feedback loop induces the p53 apoptotic function
Article Abstract:
Transactivation regulation of p53-inducible genes can be achieved through interaction between the p53 tumor suppressor gene product and the p300 transcriptional coactivator. Both p300 and p53 functions are inhibited by the adenovirus E1A protein, increasing levels of p53 accumulation and apoptosis. This process is impacted by the murine double minute 2 (mdm2) p53 transcriptional target.
