Advances in the production of human therapeutic proteins in yeasts and filamentous fungi

Article Abstract:

The challenges faced while producing the therapeutic glycoproteins and the current state of development of engineered yeast strains that are able to produce humanized glycoproteins are discussed. Yeast and fungal protein expression systems are used for the production of many industrially relevant enzymes and are extensively used to produce proteins that are not actively expressed in Escherichia coli or require glycosylation for proper folding and biological activity.

Temperature-sensitive control of protein activity by conditionally splicing inteins

Article Abstract:

A system is created which forms temperature sensitive (TS) alleles depending on the conditionally vigorous inteins. If the minimum host requirements are needed for temperature sensitive intein splicing, then this method has the capacity to create and use the conditionally vigorous inteins in various host proteins and model systems that increases the utility of temperature sensitive alleles for evaluating the functions of protein.

Enhancement of therapeutic protein in vivo activities through glycoengineering

Article Abstract:

Glycosylation analogs with new N-linked glycosylation consensus sequence introduced into the protein are screened for the presence of additional N-linked carbohydrates and retention of in vitro activity. With the help of glycoengineering the activity of proteins are prolonged, thus allowing reduced frequency of administration.
