Oxo polyene macrolide antibiotics
Article Abstract:
The oxo polyene macrolide antibiotics mycoticin, roxaticin and roflamycoin are synthesized from sodium anion of alpha-keto sulfone, optically pure beta-hydroxy ester and (S)-malic acid respectively. In mycoticin all the four 1,3-dioxane rings have chair conformations. The relative and absolute configurations in the polyol chain are identical for mycoticin and roxaticin. For roflamycoin, the syn 1,3-diol acetonides develop a chair conformation and the anti form have a twist-boat conformation. The configurations of the antibiotics are determined by analyzing the fragments of oxidative degradation.
