6-Thioguanine nucleotide accumulation in erythrocytes during azathioprine treatment for systemic connective tissue diseases: a possible index for monitoring treatment

Article Abstract:

Measuring the accumulation of 6-thioguanine nucleotide (6TGN) in red blood cells may be useful in monitoring azathioprine treatment of systemic connective tissue diseases. The drug azathioprine is frequently used to treat systemic connective tissue diseases, but response to treatment varies unexpectedly and dose adjustment guidelines are not set. 6TGNs are the major byproducts of azathioprine metabolism. Red blood cell accumulation of 6TGN was monitored in 23 patients with systemic connective tissue diseases who were taking azathioprine. Variation in red blood cell accumulation of 6TGN between individuals was high, but within the same individual, variation in 6TGN accumulation was very low. There was no significant relation between accumulation of 6TGN and the dose of azathioprine per kilogram of body weight, age of the patient, duration of treatment and incidence of toxicity.
