An induced-fit mechanism to promote peptide bond formation and exclude hydrolysis of peptidyl-tRNA

Article Abstract:

Three new structures of the large ribosomal subunit of Haloarcula marismortui complexed with peptidyl transferase substrate analogues that reveal an induced-fit mechanism in which substrates and active-site residues reposition to allow the peptidyl transferase reaction are described. The peptidyl transferase centre would use this induced-fit mechanism to help catalyse both essential reactions in which it participates.

Issues are presented concerning the benefits which can be gained for an understanding of RNA catalysis through the study of the hairpin ribozyme's crystal structure. The common features of protein enzymes and RNA are discussed.

Repopulating the RNA world

Article Abstract:

Issues are presented concerning the role played by RNA in darwinian evolution and the possibility of the existence of a RNA-like polymer during pre-RNA evolution. The properties of RNA are discussed.
