Distinct catalytic and non-catalytic roles of ARGONAUTE4 in RNA-directed DNA methylation
Article Abstract:
A study shows that ARGONAUTE4 (AGO4) binds to small RNAs including small interfering RNAs (siRNAs) originating from transposable and repetitive elements, and cleaves target RNA transcripts. The results have shown that AGO4 can recruit components that signal DNA methylation in a manner independent of its catalytic activity and its catalytic activity can be crucial for the generation of secondary siRNAs that reinforce its repressive effects.

Unlocking the potential of the human genome with RNA interference
Article Abstract:
The development of RNAi as a genetic tool in mammals is presented focusing on the advances and providing practical advice for its experimental application. RNAi pathway has emerged as a significant tool for the study of gene function in mammals and despite considerable hurdles to overcome, RNAi is expected to find a place alongside more conventional approaches in the treatment of diseases.

A resource for large-scale RNA-interference-based screens in mammals
Article Abstract:
The construction and application of a short hairpin RNA (shRNA) expression library, aiming at 9,610 human and 5,563 mouse genes, is presented. It is observed that large-scale RNAi library is likely to become a valuable resource for gene analysis and discovery.
