Identification of pathways regulating cell size and cell-cycle progression by RNAi

Article Abstract:

The effect of the RNA mediated interference (RNAi)-induced loss of function of 70% of Drosophila genes on cell-cycle progression of S2 cells was analyzed to identify genes that regulate the cell cycle to specify the size of the organism and its constituent organs. Genes that control cell size, cytokinesis, cell death and phases of the cell cycle were identified, as also a translational regulator eif-3p66, which specifically affects the Cyclin/Cdk pathway activity.

Lipoprotein particles are required for Hedgehog and Wingless signaling

Article Abstract:

A study shows that Wingless, Hedgehog and glycophosphatidylinositol-linked proteins copurify with lipoprotein particles, and co-localize with them in a developing wing epithelium of Drosophila. A novel function for lipoprotein particles, in which they act as vehicles for the movement of lipid-linked morphogens and glycophosphatidylinositol-linked proteins is proposed.

Prevalence of off-target effects in Drosophila RNA interference screens

Article Abstract:

The prevalence of 'off-target effects' (OTEs) mediated by short homology stretches within long dsRNAs in Drosophila is demonstrated. Simple measures are suggested to improve the reliability of genome-wide RNAi screens in Drosophila and other organisms.
