Structure of the multimodular endonuclease Fokl bound to DNA
Article Abstract:
FokI, a member of the bipartite restriction enzyme class, has been used to develop artificial enzymes. A crystal structure of the entire FokI enzyme bound to DNA has been determined at 2.8A resoltuion, with the enzyme found to contain amino and carboxy terminal domains, linked to DNA-recognition and cleavage functions. The cleavage domain has just a single catalytic centre and is sequestered in 'piggyback' way by the recognition domain, suggesting a new mechanism for nuclease activation.

Alternative modular polyketide synthase expression controls macrolactone structure
Article Abstract:
An alternative expression of the pikromycin polyketide synthase was found to result in the generation of two macrolactone structures. Expression in Streptomyces venezuelae generated a 14-membered ring macrolactone narbonolide. Expression of the amino-terminal truncated PikAIV resulted in skipping of the final condensation cycle.
