A NADP-glutamate dehydrogenase mutant of the petit-negative yeast Kluyveromyces lactis uses the glutamine synthetase-glutamate synthase pathway for glutamate biosynthesis
Article Abstract:
The glutamate synthesis and ammonium assimilation in the yeast Kluyveromyces lactis, involves both the glutamine synthetase (GS) and glutamate synthase (GOGAT) pathway and the NADP-glutamate dehydrogenase (GDH) pathway. The activity of GS and GOGAT in K. lactis mutants, which do not contain NADP-GDH, is more than that in the wild-type strain. The growth of these mutants is similar to that of the wild-type strain. Southern blot studies show that the NADP-GDH gene is present on chromosome VI.

Hexokinase activity is affected in mutants of Kluyveromyces lactis resistant to glucose repression
Article Abstract:
Analysis of the influence of glucose on the activities of enzymes in two different strains of Kluyveromyces lactis reveals that glucose catabolite repression significantly influences the activities of enzymes. Experimental studies on mutants of K. lactis, which do not grow on glucose, enable the identification of four types of mutants. Three of the four types of these mutants exhibit impaired hexokinase activity, revealing the vital role of hexokinase in glucose repression in K. lactis.

Research describes derivation and analysis of single and double mutants of Kluyveromyces lactis impaired or lacking in glutamate synthase (GOGAT) and GOGAT and NADP-glutamate dehydrogenase pathway respectively. Results demonstrate that glutamate biosynthesis in this bacterium requires combined action of both.
