A genetic system for the rapid isolation of aromatic-ring-hydroxylating dioxygenase activities

Article Abstract:

Research describes a genetic system that can facilitate obtaining catalytically active aromatic-ring-hydroxylating dioxygenases. Results point out that these hybrid enzymes exhibit distinct properties and can be employed to screen bacteial isolates for the enzyme activity, in complex mixtures of microorganisms or their nucleic acids such as environmental samples.

Analysis of transcription of the bph locus of Burkholderia sp. strain LB400 and evidence that the ORF0 gene product acts as a regulator of the bphA1 promoter

Article Abstract:

Transcription analysis of the bph locus of Burkholderia sp. strain LB400 indicates that the genes encoding biphenyl catabolism pathway enzymes react similarly to the presence of biphenyl, succinate, or a mixture of both. Furthermore, transcription of the genes follow either an up-regulation by growth with biphenyls or down-regulation with succinate.

Generation by a widely applicable approach of a hybrid dioxygenase showing improved oxidation of polychlorobiphenyls

Article Abstract:

A sequence-based approach is developed for the fast isolation and characterization of class II aryl-hydroxylating dioxygenase activities. The results have shown that this approach can be applied to obtain novel dioxygenase specificities that complement or supplement known ones.
