Characterization of an Escherichia coli O157:H7 plasmid O157 deletion mutant and its survival and persistence in cattle

Article Abstract:

The wild-type (ET) Escherichia coli O157:H7 and isogenic plasmid [DELTA]pO157 mutants are compared for growth rates and antibiotic susceptibilities and survival and persistence in cattle following oral challenge. The results have shown that the genes encoded on pO157 have a major impact on Escherichia coli O157:H7 metabolic activity, survival in bovine gastrointestinal tract and colonization of the bovine terminal rectum.

Viability of and plasmid retention in frozen recombinant Escherichia coli over time: A ten-year prospective study

Article Abstract:

The long-term viability and plasmid retention of recombinant Escherichia coli strains were investigated by real-time testing of master cell banks (MCBs) stored at the Roche Molecular Systems Culture Collection (RMSCC). The study shows that properly selected, frozen, and stored MCBs retain viability and maintain plasmid retention over time.

Construction and characterization of a highly efficient francisella shuttle plasmid

Article Abstract:

An enhanced shuttle plasmid that could be utilized for a variety of genetic procedures in both Francisella and Escherichia coli is constructed and characterized. The Francisella shuttle plasmid that is transformed at high efficiency is stable and does not alter the growth of Francisella in macrophages.
