Cloning of the Alcaligenes latus polyhydroxyalkanoate biosynthesis genes and use of these genes for enhanced production of poly(3-hydroxybutyrate) in Escherichia coli

Article Abstract:

A study was conducted to characterize the cloning and molecular evaluation of the Alcaligenes latus polyhydroxyalkanoates biosynthesis genes in Escherichia coli. A plasmid library of A. latus total DnA was developed by inserting A. latus genomic DNA fragments partially digested with Sau3AI into BamHI-digested PUC19. Experimental results indicated the possibility of economically producing poly(3-hydroxybutyrate) using recombinant E. coli.

Removal of endotoxin during purification of poly(3-hydroxybutyrate) from gram-negative bacteria

Article Abstract:

Poly(3-hydroxybutyrate) (PHB) with low endotoxin concentrations has been produced by chloroform extraction from gram-negative bacteria. The endotoxin level was shown to be less than 10 endotoxin units per g of PHB regardless of the bacterial strains used and the PHB content in the cell. PHB with a much lower endotoxin level could also be produced and endotoxin could be removed using the NaOH digestion approach.

Poly(3-hydroxybutyrate) production with high productivity and high polymer content by a fed-batch culture of Alcaligenes latus under nitrogen limitation

Article Abstract:

Microbiological research shows that nitrogen limitation causes the microorganism Alcaligenes latus to produce more poly(3-hydroxybutyrate) (PHB). Up to 87% PHB content is achievable under laboratory conditions with nitrogen limitation at a cell concentration of 76% dry cell weight per litre. The fed-batch culture method using DO-stat feeding is described. It could be used to produce PHB commercially.
