Conserved seed pairing, often flanked by adenosines, indicates that thousands of human genes are microRNA targets

Article Abstract:

An overrepresentation of conserved adenosines flanking the seed complementary sites in mRNAs indicates that primary sequence determinants could supplement base pairing to specify microRNA (miRNA) target recognition. In a four-genome analysis of 3' UTRs, approximately 13,000 regulatory relationships were detected above the estimate of false-positive predictions, thereby implicating as miRNA targets more than 5,300 human genes, which represented 30% of the gene set.

Prediction of mammalian microRNA targets

Article Abstract:

This article describes an algorithm-based approach to predict mammalian microRNAs to deduce microRNA:target relationship in animals. Research indicates that pairing to the 5' portion of the microRNA at the 2-8 nucleotide region seems to be the target recognition site by vertebrate microRNAs. Data suggest that the mammalian microRNA targets are enriched for genes mediating transcriptional regulation.

Passenger-strand cleavage facilitates assembly of siRNA into Ago2-cointaning RNAi enzyme complexes

Article Abstract:

Argonaute (Ago2) receives the double-stranded siRNA from the RNA-induced silencing complexes (RISC) assembly machinery directly is shown. Ago2 cleavages the siRNA passenger strand and liberates the single-stranded guide and for all siRNAs all RISC is assemsebled through the cleavage assisted mechanism.
