DNA extraction and PCR methods for the detection of Listeria monocytogenes in cold-smoked salmon

Article Abstract:

Four rapid detection protocols for detecting Listeria monocytogenes in cold-smoked salmon have been developed. Hexadecyl trimethylammonium bromide (CTAB) extraction, CTAB extraction with ether separation, CTAB extraction with column purification and CTAB extraction with particle separation and column purification can detect Listeria monocytogenes in less than 36 h via DNA extraction followed by polymerase chain reaction. They are faster than conventional detection methods and have the potential for automation.

Development of a PCR-enzyme immunoassay oligoprobe deection method for Toxoplasma gondii oocysts, incorporating PCR controls

Article Abstract:

Research describes the development of a polymerase chain reaction (PCR) amplification and detection method for Toxoplasma gondii oocyst nucleic acids. The method uses uracil-N-glycosylase to prevent false-positives, an internal control to identify false-negatives, and an oligoprobe for the detection of nucleic acids using the PCR-enzyme immunoassay.

Targeting species-specific low-affinity 16S rRNA binding sites by using peptide nucleic acids for detection of Legionellae in biofilms

Article Abstract:

The use of peptide nucleic acid (PNA) probes as a superior alternative to DNA probes, especially when used for environmental samples is described. The PNA probes performed better than the DNA probes in all experiments, in terms of both target specificity and signal intensity.
