Dipstick immunoassay to detect enterohemorrhagic Escherichia coli O157:H7 in retail ground beef

Article Abstract:

A dipstick immunoassay method was developed for detecting Escherichia coli O157:H7 in ground beef. The dipstick consisted of a strip of hydrophobic polyvinylidine difluoride-based membrane onto which capture antibodies to E. coli O157 were immobilized. The detection antibodies were specific to E. coli O157:H7. Results of dipstick reactions revealed that the method was sensitive enough to detect 0.1 colony forming unit of E. coli O157:H7 per gram of ground meat after enrichment. The procedure took 16 hours, and was specific to E. coli O157:H7.

Inactivation of enterohemorrhagic Escherichia coli in rumen content- or feces-contaminated drinking water for cattle

Article Abstract:

The different treatments to eliminate or control Escherichia coli O157:H7 in the presence of rumen content or feces-contaminated drinking water are described. These treatments should be applied periodically to drinking water troughs and then flushed, rather than being added continuously, to avoid reduced water consumption by cattle.

Construction and characterization of shuttle vectors for succinic acid-producing rumen bacteria

Article Abstract:

Various shuttle vectors having the origins of replication functioning in Escherichia coli and rumen bacteria are constructed and characterized. These shuttle vectors can also be used for genetic and metabolic engineering of the rumen bacteria that produce succinic acid.
