Effect of introns and AT-rich sequences on expression of the bacterial hygromycin B resistance gene in the basidiomycete Schizophyllum commune

Article Abstract:

A model is presented demonstrating that truncation of transcripts and intron-dependent mRNA accumulation are two separate events that influence expression of the hygromycin B resistance gene in the bacterium Schizophyllum commune.

Highly efficient production of laccase by the basidiomyce Pycnoporus cinnabarinus

Article Abstract:

A transformation and expression system for pycnoporus cinnabarinus was developed and used to produce high levels of the homologous laccase lac1. The results show that pycnoponus cinnabarius may be a host of choice for the production of other proteins as well.

RNA-medicated gene silencing in monokaryons and dikaryons of Schizophyllum commune

Article Abstract:

A hairpin construct of SC15 was made to ensure gene silencing to determine whether a mechanism similar to RNAi is operating in Schizophyllum commune. RNA-based gene silencing occurs at a much higher frequency than homologous recombination and enables silencing of functional genes in both nuclei of a dikaryon by the expression of a hairpin construct in only one of the nuclei.
