Engineered biosynthesis of a novel amidated polyketide, using the malonamyl-specific initiation module from the oxytetracycline polyketide synthase

Article Abstract:

The gene cluster of oxytetracycline (oxy and otc genes) polyketide synthases (PKSs) genes from Streptomyces rimosus was sequenced. The sequence analysis revealed a total of 21 genes between the ortA and ortB resistance genes while the in vivo reconstitution using strain Ch999 revealed that the minimal PKS and oxyD are necessary and sufficient for the biosynthesis of amidated polyketides.

Characterization of the sporulation control protein SsgA by use of an efficient method to create and screen random mutant libraries in streptomycetes

Article Abstract:

A novel and efficient method is described to create, propagate and screen a large collection of random ssgA mutants streptomycetes. The three-dimensional structures of the SsgA protein might help in understanding the role of SsgA in the control of the growth and development of streptomycetes.

Efficient synthesis of simvastatin by use of whose-cell biocatalysis

Article Abstract:

A one-step, whole-cell biocatalytic process is developed for synthesizing simvastatin from monacolin J. The whole-cell biocatalysis is found to be an efficient alternative to the multistep semisynthetic transformations.
