Fluorescence in situ hybridization using 16S rRNA-targeted oligonucleotides reveals localization of methanogens and selected uncultured bacteria in mesophilic and thermophilic sludge granules

Article Abstract:

The locations of several microorganisms in mesophilic and thermophilic granular sludges have been identified by using a combination of the in situ hybridization technique with confocal laser scanning microscopy. The sludges are contained in upflow anaerobic sludge blanket reactors with sucrose-, acetate-, and propionate-based artificial wastewater. Results show that Methanosaeta-, Methanobacterium-, Methanospirillium-, and Methanosarcina-like cells were detected by the oligonucleotide probes and were found to be localized inside the granules.

Application of pseudomourein endoisopeptidase to fluorescence in situ hybridization of methanogens within the family Methanobacteriaceae

Article Abstract:

A study applied pseudomurein endoisopeptidase (Pei) to prior to 16S rRNA-targeting fluorescence in situ hybridization (FISH) analysis for clearly visualizing the methanogens in the family Methanobacteriaceae. Results reveal that Pei treatment is effective for FISH analysis of methanogens that reveal impermeability to the probe.

To detect a specific group of microorganisms in complex ecosystems, a simple approach is designed. This approach consists of the sequence specific cleavage of rRNA molecules with oligonucleotides.
