Monte Carlo pedigree disequilibrium test for markers on the X chromosome

Article Abstract:

A disequilibrium test applicable to pedigrees and X-chromosomal markers is proposed and is extended to accommodate missing genotypes through a Monte Carlo (MC) estimation procedure. It is observed that the MC version of the X-chromosomal pedigree disequilibrium test is a valid test for association under linkage with the assumption that the pedigrees and their associated affection patterns are drawn randomly from a population of pedigrees with at least one affected offspring.

Genome screens using linkage disequilibrium tests: optimal marker characteristics and feasibility

Article Abstract:

Optimal marker characteristics and feasibility in genome screens using linkage disequilibrium (LD) tests are discussed with crucial information for designing effective screening studies for markers that show LD with a disease locus, comparing diallelic markers to multiallelic markers. Findings indicate use of multiallelic markers is the better choice in almost all cases.

Disequilibrium likelihoods for fine-scale mapping of a rare allele

Article Abstract:

A likelihood-based method of fine-scale mapping of a rare monophyletic disease is presented and applied, as an example, to the Werner syndrome in the Japanese. The method depends on a coalescent model of the ancestry of a sample of disease alleles and involves only disequilibrium mapping for a rare allele that is monophyletic in origin.
