Ribotyping to compare Fusobacterium necrophorum isolates from bovine liver abscesses, ruminal walls, and ruminal contents

Article Abstract:

A research was conducted to genetically compare Fusobacterium necrophorum isolates from multiple abscesses of the same liver and isolates from liver abscesses, the ruminal wall, and ruminal contents from the same animal using restriction fragment length polymorphism analysis of rRNA genes from Escherichia coli. Random priming was utilized to transcribe a nonradioactive labeling system from a mixture of 16S and 23S rRNAs from E. coli MRE600. Results showed that ribotyping was useful for comparing various strains of F. necrophorum and for determining the strains that cause liver abscesses.

PCR-based DNA amplification and presumptive detection of Escherichia coli O157:H7 with an internal fluorogenic probe and the 5' nuclease (TaqMan) assay

Article Abstract:

Research was conducted to examine the development and evaluation of the specificity and sensitivity of a PCR-based 5' nuclease assay for presumptively detecting Escherichia coli O157:H7 DNA. Data indicate that incorporating preenrichment and secondary enrichment steps and an IMS recovery process could further improve the sensitivity of the detection procedure. Results suggest that presumptive pathogen detection systems using 5' nuclease assay components can be integrated readily into standard culture/detection methods to reduce required time for presumptive detection.
